Presence of HBV S-gene mutants in immunocompromised patients.

چکیده
Hepatitis B virus (HBV) is responsible for a serious, potentially fatal disease that affects approximately two billion people worldwide. Although an extended HBV immunization program has run since 1991, representing the most effective preventive measure possible, leading to a dramatic reduction of HBV hepatitis incidence, 4.5 million new HBV infections still occur every year. In 1988 the emergence of HBV S-gene mutants was first observed in Italian vaccinated children’s sera with the presence of both HBs antigen and anti-HBs antibodies. S-gene sequence revealed glycine (G) to arginine (A) substitution at position 145, within the a-determinant of S-gene, causing conformational changes that allow for the virus to escape the vaccine-induced response (Zanetti, 1988). Other similar mutations were later found in immunocompromised patients, responsible for HBV reactivation by immune escape in previously anti-HBs immune persons (Sheldon, 2008). As the prevalence of HBV surface antigen (HBsAg) variants is unknownwe investigated the presence of HBsAg mutations in 200 consecutive Ligurian patients, monitored for viral load and drug resistance. Virus sequences were obtained by the Trugene®

YMDD and FLLAQ Mutations in Chronic Hepatitis B Patients
Abstract Background and Objective: Lamivudine is the first orally available drug approved for treatment of chronic hepatitis B. Mutations at the YMDD and FLLAQ motifs in the domains of HBV polymerase gene contribute resistance to lamivudine. This study was aimed to determine the rate of YMDD and FLLAQ mutants in hepatitis B patients in Golestan Province, Iran. Material and methods: In this cros...
متن کاملIdentification of Drug Resistant Mutants of HBV (Hepatitis B Virus) by Direct Sequencing in Iranian Patients Treated with Lamivudine
Background and Aims: lamivudine is amongst the antiviral for drug chronic hepatitis B treatment. During therapy with lamivudine, variants may emerge with YMDD mutation in the reverse transcriptase (RT) region of polymerase gene. This mutation might have a role in drug resistant for HBV. Materials and Methods: HBV DNA extraction from serum sample of 88 patients, were subjected to nested PCR for ...
متن کاملPrevalence of Hepatitis B Virus, Genotypes, and Mutants in HBsAg-Positive Patients in Meerut, India
Background: Genetic changeability of hepatitis B virus (HBV) signifies a challenge for the sensitivity of immunologic and molecular diagnostics. Therefore, knowing the spread of HBV genotypes (GENs) and mutation has considerable impacts on treatment strategies, vaccination program, diagnosis, and prevention. The present study aimed to detect HBV GENs and mutants in HBsAg-positive patients. Meth...
متن کاملImmunocompromised patients with HBsAg a determinant mutants: comparison of HBsAg diagnostic assays.
Hepatitis B surface antigen (HBsAg) is considered the best marker for the diagnosis of hepatitis B virus (HBV) infection. Mutations of the s gene involving amino acid substitutions within the a determinant could affect the sensitivity of diagnostic tests. In the present study, HBsAg mutants were detected in 3 immunocompromised patients, previously found to be HBsAg negative and anti-HBs positiv...
متن کاملCorrelation between Polymorphism of -56 SNP (T/C) Interferon-γ Receptor 1 Gene and Chronic HBV Infection
Background and Aims: chronic HBV infection is one of the most common viral infections in worldwide which many factors such as genetic factors are involved in pathogenesis of disease. Gamma interferon (IFN-γ) and its receptor (INFGR) play a critical role in the immune response to HBV infection. Single nucleotide polymorphisms (SNPs) are effective on level of gene expression, The aim of thi...
